The German side broke the deadlock in the 36th minute, after a precise long ball from Nico Schlotterbeck, found Fullkrug, who timed his run perfectly, controlled the ball before slotting home past Donnarumma.
The visitors came close to levelling the score early in the second half, with Kylian Mbappe hitting the post and in the rebound Achraf Hakimi struck the woodwork again.
Dortmund's response was thwarted by missed opportunities from Julian Brandt and Fullkrug.
PSG continued to push forward, with Mbappe and substitute Randal Kolo Muani testing Dortmund's defence, but the hosts held firm, with Gregor Kobel making crucial saves.
Despite late chances for both teams, Dortmund secured the victory and will carry their lead to Paris for the second leg next week.
